IR Laser Diode Imaging

 

An Laser Analytics IR diode laser, tunable between about 1530-1620 cm-1, has been coupled to a Spectra-Tech Research IRplan microscope (15x objective, 10x condenser) equipped with a 0.25 mm2 broad-band MCT detector element and a Cell Robotics programmable microscope stage. The FTIR instrument is also used to calibrate the wavelength output from the diode laser by passing the IR beam through the interferometer to the internal detector. Light from the IR diode laser is collimated by an off-axis parabolic mirror and passed through a chopper before entering the microscope. The transmitted light is collected and focussed onto the detector. The modulated signal is then passed to a lock-in amplifier, digitized, and passed to the computer.
